[摘要] 目的 研究瑞香狼毒水提取物对人膀胱癌T24细胞的抑制作用及生存素(Survivin)表达的影响。 方法 用不同浓度的瑞香狼毒水提取物处理T24细胞，分别用MTT法检测膀胱癌细胞抑制率，流式细胞术检测细胞凋亡率，免疫细胞化学检测药物作用后细胞中Survivin蛋白的表达。 结果 瑞香狼毒水提取物干预细胞后，①膀胱癌T24细胞增殖受到抑制，抑制效应呈剂量依赖性并随时间的延长逐渐升高；②瑞香狼毒水提取物可以促进膀胱癌T24细胞的凋亡；③膀胱癌T24细胞中的Survivin蛋白表达下降。 结论 瑞香狼毒水提取物可以通过下调Survivin蛋白的表达从而抑制膀胱癌T24细胞增殖，促进其凋亡，并具有浓度和时间依赖性。

The effects of the extracts from Stellera chamaejasme L. on expression of survivin gene of human bladder cancer T24 cell

[Abstract] Objective To study the inhibition effects and the expression of survivin gene of the extracts from Stellera chamaejasme L. on human bladder cancer T24 cells. Methods The T24 cells were dealt with different concentrations of extracts from Stellera chamaejasme L.. MTT, flow cytometry and immunocytochemical staining were used to investigate the suppression ratio of bladder cancer cells, the apoptosis rate and the expression of survivin protein in cells respectively. Results After intervention of the extracts from Stellera chamaejasme L., ①bladder cancer T24 cells proliferation was inhibited, the inhibitory effects were in a dose-dependent manner and increased gradually with the extend of time; ②the extracts from Stellera chamaejasme L. could induce apoptosis of bladder cancer T24 cells; ③the survivin protein expression of Bladder cancer T24 cells was decreased. Conclusion The extracts from Stellera chamaejasme L. can down-regulate the expression of survivin protein to inhibit the proliferation of bladder cancer T24 cells and to promote their apoptosis, and is in a time and concentration-dependent manner.
